One unit is defined as the amount of enzyme required to completely degrade 1μg of DNA in 10 minutes at 37°C in 50μl of a buffer containing 40mM Tris-HCl (pH 7.9 at 25°C), 10mM NaCl, 6mM MgCl2, 10mM CaCl2.
